The deep well microplates are commonly used for sample storage in life science laboratories. There is an important aspect associated with the manufacture of these plates, which is the selection of clean raw materials for injection moulding.

They are made of polypropylene, which is an inert and heat resistant material, ideal for sample storage. However, all grades of polypropylene are not the same; for example, injection moulding grades of polypropylene often have high concentrations of chemicals to aid the moulding process. This allows rapid production of plates and lower costs, but may compromise the integrity of samples or compounds which are stored in such plates for extended periods.

With regards to one of the microplates, the 96 deep well plate was designed so that it would make the interchange of plates simpler in automated systems. So, for applications using the GenoGrinder™ and TissueLyzer™, a specially reinforced version of this plate is available with extractable-free, medical-grade molecules called polymer.

The microplate 96 well is specifically for seed and leaf genomic studies, which is an area within genetics that focuses on the analysis of an organism. The microplate 96 well is also:

These microplates are made in virgin polypropylene to minimise extractable. Their maximum volume is 2.075 ml per well, and working volume is a useful 1.85 ml, more than any other comparable ‘2 ml Round Well’ plate in this class.

There is a design called the ‘common wall’ which allows the highest possible volume to be used while maintaining an overall height of just 45 mm, complete with ANSI/SLAS standard base and footprint.

The plates are manufactured in proper room conditions, and a significant number are DNase/RNase free. They are mainly made from polypropylene, which is inert material giving heat and solvent resistant qualities.

Porvair Sciences offers a wide range of products to implement SPE extraction. Solid-phase extraction (SPE) is a method of sample preparation that concentrates and purifies analytes from solution by sorption, followed by elution of the analyte with a solvent appropriate for instrumental analysis, such as LC-MS.

MicroLute™ is the original SPE 96-well plate format for SPE. According to Porvair Sciences, the device consists of a ‘polypropylene 96-well plate loaded with a choice of sorbent and sorbent volumes’. Also, they mention that the frits have a ‘mean pore size of 20 microns and are made from microporous high-density polyethylene’.

Solid-phase extraction uses the difference of attraction between an analyte and interferents, present in a liquid matrix, for a solid phase (sorbent). It is the attraction that allows the separation of the target analyte from the interferents.

There are several steps involved with a solid-phase extraction; they include:

First, the cartridge is conditioned with a solvent to wet the sorbent. After that, the loading solution containing the analyte is infiltrated through the solid phase (SP). Ideally, the analyte and some impurities are retained on the sorbent. The sorbent is then washed to remove impurities, and the analyte is collected during this elution step.

Their selectivity is limited, and the target analyte often co-elutes with interfering compounds. On the other hand, there are molecularly imprinted polymers are the most highly selective phases available today. This technology enables to reduce the presence of interferents during the elution step and obtain higher recoveries in chromatography analysis even at low levels.
